Abstract

Background Searching and identifying the role of effective factors in coronary artery disease (CAD), such as inflammatory factors and cytokines, are important for increasing our understanding of the disease pathophysiology. The nerve growth factor (NGF) is one of risk factors of CAD that plays a key role in expressing and controlling inflammatory responses by macrophages. Objective This study investigated the relationships between NGF promoter polymorphism, − 198 from the transcription start site, and NGF gene expression in patients with CAD and healthy subjects in Iranian population. Methods After coronary angiography, 302 patients with > 70 vessel stenosis were selected as CAD group; and 310 normal angiogram subjects were selected as control group. Genomic DNA and total RNA were extracted from peripheral whole blood and polymorphism and expression of NGF gene was determined by SSP-PCR and real-time PCR. Results There was significant association between age, gender, smoking status, and diabetes mellitus with CAD disease (p < 0.001). T allele's frequency among CAD and control groups was 54.1 and 46.8, respectively, OR (95%CI): 1.34 (1.07–1.68), p = 0.010. Expression of this gene in the case group was reduced significantly compared to the control group (p < 0.05). The genotype TT and allele T were significantly associated with CAD. Conclusion According to our results, the allele T of rs11102930 has a significant association with CAD progression. This polymorphism may be involved in regulation of NGF expression and plaque development in atherosclerosis. © 2017 Elsevier B.V.
